糖蛋白组学是研究蛋白质糖基化的一种科学,糖基化是蛋白质的一种重要翻译后修饰,它在细胞的各种生理和病理过程中扮演着关键角色。传统的糖蛋白质组学依赖于液相色谱与串联质谱联用技术,但这些技术在确定聚糖结构方面存在挑战。

为了解决这一问题,复旦大学的研究人员提出了DeepGP框架,这是一种基于Transformer和图神经网络的混合深度学习框架。DeepGP框架能够预测糖肽的MS/MS光谱和保留时间(RT),这对于糖肽的结构鉴定至关重要。两个图神经网络模块分别用于捕获分支糖结构和预测糖离子强度,而预训练策略则缓解了糖蛋白质组学数据不足的问题。

DeepGP框架的提出,不仅能够提高糖肽鉴定的灵敏度,还能够通过结合现有的糖肽搜索方法(如pGlyco3),实现对相似聚糖的有效区分。这一技术的突破将为医学研究、生物制药以及疾病诊断等领域带来深远的影响。
